Evaluation and optimization of model systems for spermatogenesis, such as testis grafting, spermatogonial transplantation, and in vitro spermatogenesis, are often limited in domestic animals by a lack of molecular and immunocytochemical markers for germ cell progression. The first objective of this study was to characterize the histological progression of germ cells and correlative stage-specific gene expression during the establishment of spermatogenesis in the Holstein bull. Bulls were castrated at 4 wk intervals between 8 and 16 wk postnatally and 2 wk intervals thereafter through 32 wk. Testis development was correlated more highly with bull weight than age. Expression of TP1 and PRM2 coincided with the histological appearance of spermatids and the ratio of the isoforms of SCF from Sertoli cells changed with age. Microarray analysis of testis tissue at 8, 20, and 32 wk showed that 50% of the bovine genome is expressed at some point during prepubertal development, with 21% of these genes being differentially expressed between the ages examined.
